Embryonic kidney HEK-293
cell line / Homo sapiens / embryonic kidney
- Animal species:
- Human, Homo sapiens.
- Origin:
- embryonic kidney
- Age:
- human embryo
- Tissue:
- embryonic kidney
- Morphology:
- epithelial, adherent
- Tumoroids obtained:
- yes
- Subcutaneous growth:
- yes
- Molecular profile:
- Vitronectin
- Tumorigenicity:
- yes, tumors in nude mice
- Karyology:
- hypotriploid human cell line. The modal chromosome number was 64, occurring in 30% of cells. The proportion of cells with higher ploidy was 4.2 %. Der(1)t(1.15) (q42.q13), der(19)t(3.19) (q12.q13), der(12)t(8.12) (q22.p13) and four other marker chromosomes were common to most cells. Five other markers were found only in some cells. Markers derembryonic kidney(1) and M8 (or Xq+) were often paired. There were four copies of N17 and N22. Notably, in addition to three copies of the X chromosome in most cells, there were paired Xq+ and single Xp+.
- Contamination control:
- not detected
- Viral infection:
- although in an earlier study the cells contained adenovirus 5 DNA from the right and left ends of the viral genome. It is now known that it contains only left-end sequences.
- Species identity control:
- karyological, isoenzyme (LDH and G6PD) and immunofluorescence analysis.
- Application area:
- biotechnology, toxicology, screening, testing of transfection efficiency, virucidity test for host virucidity
- Culture method:
- in vitro partially adhesion, in vivo (see Appendix 1).
- In vitro culture conditions:
- medium – EMEM fetal bovine serum 10%. passaging procedure - cell detachment using trypsin 0.25 %: Versene 0.02 % (1:3), split ratio 1:3 -1:5 cryopreservation – growth medium 65%, 5% DMSO, 30% fetal bovine serum It is normal for these cells to recover slowly after cryopreservation, and they may need 7-14 days to start growing actively.(see Appendix 1).
- Physical state of the sample:
- frozen.
- Cryopreservation:
- liquid phase and nitrogen vapor (see Appendix 1).
- Viability after cryopreservation:
- 94% (trypan blue staining after thawing).
- Collections:
- A.P. Avtsyn Research Institute of Human Morphology, Federal State Budgetary Scientific Institution "B.V. Petrovsky Russian Scientific Center of Surgery", ATCC.
- Patent, certificate:
- no
- Note:
- transfection is possible. Studies have shown that HEK 293 have more properties of undifferentiated neurons. A comparison of the HEK 293 transcriptome with transcriptomes of kidney, adrenal and nerve cells determined that HEK 293 cells are most similar to adrenal cells, which in turn also have characteristics of undifferentiated neurons.
- Intended use:
- these samples are intended for laboratory research only.
